CDKN2B encodes p15INK4b, a cyclin-dependent kinase inhibitor that binds and inhibits CDK4 and CDK6 to enforce cell-cycle arrest at the G1 checkpoint, acting as a tumor suppressor. The CDKN2B/CDKN2A region on chromosome 9p21 is one of the most consistently replicated genome-wide association loci for primary open-angle glaucoma (as well as coronary artery disease and type 2 diabetes), with CDKN2B implicated in trabecular-meshwork cell senescence and increased outflow resistance that raise glaucoma risk.
