EDA2R encodes ectodysplasin A2 receptor, a tumor necrosis factor receptor superfamily member on the X chromosome that binds the EDA-A2 ligand and activates JNK and NF-kB signaling to promote apoptosis. In hair follicle dermal papilla cells, EDA2R is upregulated in balding scalp tissue and its signaling induces DKK1, which suppresses hair growth, and the missense variant rs1385699 is one of the strongest and most replicated genetic markers for male-pattern hair loss.
