SLCO1B1

SLCO1B1 encodes OATP1B1, a transporter on the liver-facing (basolateral) membrane of hepatocytes that pulls drugs, including statins, out of the bloodstream and into the liver for metabolism and clearance. Reduced-function SLCO1B1 variants slow this hepatic uptake, raising circulating statin concentrations and increasing the risk of statin-associated muscle symptoms, most notably with simvastatin.
