rs9923231 is the common VKORC1 -1639G>A promoter variant used in warfarin dosing algorithms. On the GRCh38 forward strand it is C>T. Each T copy is linked to lower VKORC1 expression, progressively greater warfarin sensitivity, and a lower average dose requirement. It is one input alongside CYP2C9, clinical factors, and INR monitoring; it cannot determine a dose by itself.
