F2 encodes prothrombin, a vitamin K-dependent plasma protein synthesized in the liver that is cleaved to form thrombin, the central enzyme of the blood coagulation cascade that converts fibrinogen to fibrin and activates platelets. The promoter variant rs1799963 (prothrombin G20210A) increases prothrombin levels and is a well-established hereditary risk factor for venous thromboembolism, including deep vein thrombosis and pulmonary embolism.
