NOD2
NOD2 encodes an intracellular pattern-recognition receptor expressed in monocytes, macrophages, and intestinal Paneth cells that detects bacterial peptidoglycan fragments (muramyl dipeptide) and activates NF-kB-mediated innate immune signaling. Loss-of-function variants (including rs2066844, rs2066845, and rs2066847) impair bacterial sensing and mucosal barrier defense in the gut and are the strongest known genetic risk factors for Crohn's disease, a form of inflammatory bowel disease.
