rs10305420
rs10305420 is a missense variant in GLP1R, c.20C>T causing Pro7Leu near the receptor's N-terminal signal peptide. In a 2026 study of 27,885 people taking semaglutide or tirzepatide, each T allele was associated with about 0.76 kg more weight loss and more nausea or vomiting. The weight-loss result was independently replicated in 4,855 All of Us participants; the side-effect result has not yet been independently replicated. The same allele points to both effects, so they must be presented together.
