rs1360780

rs1360780 is an intronic variant in FKBP5, a co-chaperone that regulates glucocorticoid receptor sensitivity and negative feedback within the HPA stress axis. The T allele is associated with greater stress-induced FKBP5 mRNA induction and, particularly in people with childhood trauma exposure, with DNA demethylation at nearby glucocorticoid response elements in intron 7 that further amplifies FKBP5 expression and blunts cortisol feedback. It is used as genetic context for stress-response/cortisol-signaling pathway function and has been associated in gene-by-environment studies with PTSD risk and altered response to trauma-focused psychotherapy.
