rs1800437 is the GIPR Glu354Gln (E354Q) missense variant in the glucose-dependent insulinotropic polypeptide receptor gene. The Gln354 allele changes long-term GIP receptor signaling and trafficking. In one 2026 study it was associated with more nausea and vomiting in people taking tirzepatide, with no association in semaglutide users and no weight-loss effect. The drug-side-effect association has not been independently replicated, and its mechanism remains unresolved because cell studies found that tirzepatide activates both receptor versions similarly.
