rs2231142 is the ABCG2 Gln141Lys (Q141K) missense variant, a common coding change in the ABCG2 urate transporter gene. The Lys141 protein folds and inserts into the membrane less efficiently and is degraded faster, cutting extra-renal (gut) urate excretion and raising serum uric acid. Each copy of the risk (T) allele raises serum urate and roughly 1.5-2x's gout risk, making it one of the two strongest common variants (with SLC2A9) for hyperuricemia and gout.
